Recent clinical data and real-world studies are revealing that GLP-1 medications (such as semaglutide and tirzepatide) offer significant, measurable benefits for mental health, particularly for depression and anxiety.
Here is what the latest research tells us about how GLP-1s are impacting depression:
Significant Reductions in Psychiatric Hospitalizations
A massive registry-based study out of Sweden tracking nearly 100,000 individuals over a period of 13 to 14 years found that people using GLP-1 medications had significantly fewer psychiatric hospital visits and took less time off work due to mental health issues. During periods when patients were taking semaglutide, the risk of depression was 44% lower, and the risk for anxiety disorders dropped by 38%. The medications were also linked to a 47% reduction in substance use disorders and a reduced risk of suicidal behavior.
Improved Motivation and Reward Processing
One of the core struggles in Major Depressive Disorder (MDD) is a lack of motivation and anhedonia (the inability to feel pleasure). A recent clinical trial found that treating MDD patients with semaglutide specifically improved their motivation and reward-related dysfunction. Computational modeling showed that the medication reduced “effort discounting,” meaning it made patients more willing to exert physical effort to achieve a reward by reducing the perceived cost of that effort.
The Neurobiological Mechanisms

Reversal of Safety Concerns
You may have previously heard fears that GLP-1s could cause depression. When these drugs first gained popularity, the FDA received adverse event reports suggesting they might trigger suicidal thoughts or self-harm. However, subsequent large-scale research explicitly disproved this, showing no increased risk and, in fact, a decreased risk of worsening mental health symptoms. Because the evidence so strongly points to GLP-1s being safe and potentially protective for mental health, the FDA actually requested the removal of suicidal behavior warnings from the labels of certain GLP-1 medications like Wegovy.
